چکیده

Ovarian carcinoma is one of the leading causes of cancer-related death among women. The purpose of this study was to investigate the frequency of CD4+CD25+CD127regulatory T cells (Tregs) in the peripheral blood of patients with epithelial ovarian cancer (EOC) and to explore their immune regulatory roles. 118 female patients with EOC and 30 healthy women were recruited. The percentage of CD4+CD25+CD127Tregs within the CD4+ T cell population in the peripheral blood was detected by flow cytometry. The relation between clinicopathologic factors and Tregs frequency was analyzed. To study the immunoregulatory capacity of CD4+CD25+CD127Tregs, cytokines produced by CD4+CD25+CD127Tregs was measured by ELISA and the proliferation of cells was measured by incorporation of [3H] thymidine. The percentage of CD4+CD25+CD127Tregs within the CD4+ T cell population in the peripheral blood of EOC patients was 5.34 ± 1.77%, significantly higher than that in healthy women. No correlation of Tregs frequency with tumor stage, differentiation grade and histological subtype was found. CD4+CD25+CD127Tregs derived from peripheral blood did not produce IFN-γ, but large amounts of IL-10. The isolated CD4+CD25+CD127Tregs notably decreased the proliferation of CD4+CD25T cells. We provide evidence of increased frequency of Tregs with potent immunosuppressive features in the peripheral blood of patients with EOC, which may be responsible for immune tolerance in EOC.

FoxP3+ Regulatory T Cells in Peripheral Blood of Patients with Epithelial Ovarian Cancer

Background: Ovarian cancer is the fifth leading cause of death from malignancy in women. CD4 +CD25+FoxP3+ regulatory T (Treg) cells are a subset of T lymphocytes with great inhibitory impact on immune response. Objectives: To investigate the percentage of CD4 +CD25+FoxP3+ regulatory T cells in the peripheral blood of the Iranian patients with epithelial ovarian cancer compared to healthy women ...

Concomitant Increase of OX40 and FOXP3 Transcripts in Peripheral Blood of Patients with Breast Cancer

Background: Regulatory T cells (T-regs) have an important role in cancer by suppression of protective antitumor immune responses. Regulatory T cells express the forkhead/winged helix transcription factor (FOXP3) and OX40 molecules which have important regulatory roles in the immune system. Objective: To evaluate FOXP3 and OX40 transcripts in the peripheral blood mononuclear cells of women with ...

Regulatory T Cells in Colorectal Cancer

One of the essential protection mechanisms of immune system is Treg cells which play an important role in maintenance ofimmune homeostasis. However, they may also inhibit immune functions against tumor cells. It has been reported that Treg cellsare increased in patients suffering from different types of cancer. Increased number of Treg cells has been shown in tumor lymphnodes, peripheral blood,...

Regulatory T Cells and Myeloid-Derived Suppressor Cells in Patients with Peptic Ulcer and Gastric Cancer

Background: Regulatory T Cells (Tregs) and Myeloid-Derived Suppressor Cells (MDSCs) are two main regulatory cells modulating the immune responses in inflammation and cancer. Objective: To investigate and compare Tregs and MDSCs in peptic ulcer and gastric cancer. Methods: Patients with dyspepsia were selected and divided into three groups of non-ulcer dyspepsia (NUD, n=22), peptic ulcer disease...

Altered Suppressor Function of Regulatory T Cells in Type 1 Diabetes

Background: Type 1 diabetes (T1D) is a T cell mediated autoimmune disease targeting the insulin-producing β cells within pancreatic islets. Autoimmune diseases may develop as a consequence of altered balance between regulatory (Tregs) and autoreactive T cells. Objectives: To evaluate Treg cells frequency and suppressive function in the peripheral blood of newly diagnosed T1D patients in compari...
